Norway maple and sycamore, two Acer genus species, have an important ecological value and different sensitivity to stressing factors being currently aggravated by climate change. Seedling growth is postulated to be the main barrier for successful plant establishment under the climate change scenarios. Therefore, the differences in redox regulation during the seedling performance of Norway maple and sycamore were investigated. Seeds of the two Acer species exhibited an identical high germination capacity, whereas seedling emergence was higher in sycamores. PCA analyses revealed that there is more diversification in the leaf characteristics than roots. Norway maple displayed a higher chlorophyll content index (CCI) with a similar leaf mass whereas sycamore seedlings exhibited a higher normalized difference vegetation index (NDVI), higher water content, higher root biomass and higher shoot height. Based on NDVI, sycamore seedlings appeared as very healthy plants, whereas Norway maple seedlings displayed a moderate healthy phenotype. Therefore, redox basis of seedling performance was investigated. The total pool of glutathione was four times higher in sycamore leaves than in Norway maple leaves and was reflected in highly reduced half-cell reduction potential of glutathione. Sycamore leaves contained more ascorbate because the content of its reduced form (AsA) was twice as high as in Norway maple. Therefore, the AsA/DHA ratio was balanced in sycamore leaves, reaching 1, and was halved in Norway maple leaves. Nicotinamide adenine dinucleotide phosphate content was twice as high in sycamore leaves than in Norway maples; however, its reduced form (NADPH) was predominant in Norway maple seedlings. Norway maple leaves exhibited the highest anabolic and catabolic redox charge. The higher reduction capacity and the activity of NADPH-dependent reductases in Norway maple leaves possibly resulted in higher CCI, whereas the larger root system contributed to higher NDVI in sycamore. The different methods of controlling redox parameters in Acer seedlings grown at controlled conditions provided here can be useful in understanding how tree species can cope with a changing environment in the future.

Previous studies have shown that women perceive male faces with a more reactive immune system as more attractive, but whether body odor might likewise provide cues to immune function has not been investigated yet. These two studies tested a possible relationship between body odor quality and immunoreactivity (Study 1) and immune system function (Study 2). In Study 1, we collected body odor samples from 21 men just before and two weeks after vaccination against hepatitis A/B and meningococcus. We determined the levels of specific antibodies (selected as markers of immune system's reactivity), testosterone, and cortisol. Subsequently, 88 female raters assessed the odor samples for their attractiveness, intensity, and healthiness. In Study 2, we collected body odor and blood samples from 35 men and women. We assessed key parameters of their innate and adaptive immunity, such as complement activity or total lymphocyte T and B counts and asked 95 raters to assess the odor samples for their attractiveness, intensity, and healthiness. In Study 1, we found no significant association between antibody levels induced by vaccination and perceived body odor attractiveness, intensity, and healthiness. We also found no significant relationship between antibody levels and steroid hormones (testosterone and cortisol). In Study 2, we likewise found no association between basal key parameters (innate and adaptive) of the immune system and body odor quality. Our results indicate that body odor may not serve as a cue to the reactivity of the immune system.

Masculinity-related morphological traits are supposed to be honest indicators of a man's biological quality. While some studies showed that sexually dimorphic traits are related to various aspects of biological condition such as general health, immunity or fertility, still little is known about the relationship between masculine traits and the effectiveness of innate and adaptive immunity in humans. The aim of this study was to see if masculine traits, which are dependent on androgen levels in foetal and pubertal stages of development, are related to the immune quality in healthy men. The immune quality was evaluated for 91 healthy men aged 19-36 years. Immunity measurements included innate and adaptive parameters. General health status, age, testosterone level, BMI, physical activity, and smoking were controlled. The shoulder-to-hip ratio (SHR), 2D:4D digit ratio and hand-grip strength (HGS) were used as markers of masculinization. The regressions showed that when controlling for confounds, masculinity-related traits were in general not related to innate and adaptive immunity. Only a weak association was observed for right 2D:4D ratio and T-lymphocyte counts (but it becomes non-significant after adjustment for multiple comparisons). Our results do not support the premise that masculinity is a cue for immunological quality in men. However, the positive association between right 2D:4D and T lymphocytes might suggest that further studies are needed to verify if androgen stimulation in prenatal development might be related to immunity in adulthood.

Temperature is a key environmental factor restricting seed germination. Rose (Rosa canina L.) seeds are characterized by physical/physiological dormancy, which is broken during warm, followed by cold stratification. Exposing pretreated seeds to 20 °C resulted in the induction of secondary dormancy. The aim of this study was to identify and functionally characterize the proteins associated with dormancy control of rose seeds. Proteins from primary dormant, after warm and cold stratification (nondormant), and secondary dormant seeds were analyzed using 2-D electrophoresis. Proteins that varied in abundance were identified by mass spectrometry. Results showed that cold stratifications affected the variability of the highest number of spots, and there were more common spots with secondary dormancy than with warm stratification. The increase of mitochondrial proteins and actin during dormancy breaking suggests changes in cell functioning and seed preparation to germination. Secondary dormant seeds were characterized by low levels of legumin, metabolic enzymes, and actin, suggesting the consumption of storage materials, a decrease in metabolic activity, and cell elongation. Breaking the dormancy of rose seeds increased the abundance of cellular and metabolic proteins that promote germination. Induction of secondary dormancy caused a decrease in these proteins and germination arrest.

OBJECTIVES: More symmetric organisms are perceived as more attractive. Fluctuating asymmetry (FA) i.e. small, random deviations from perfect bilateral symmetry, is supposed to inform about developmental instability. According to the good genes hypothesis, a low level of FA is a putative cue to an organism's biological quality. An important aspect of this quality is the immune system functioning. The aim of this study was to evaluate the relationship between immune system functioning and body symmetry in healthy people. MATERIALS AND METHODS: The composite body FA (cFA) was assessed on the basis of six bilateral traits (on hands and feet). The ISF was determined by many innate (total complement and lysozyme activity, neutrophils function) and adaptive immune parameters (T CD3 and B CD19 lymphocytes, total IgA and IgG and response to flu vaccine). A total of 98 men and 92 women were subjected to flu (among them 37 men and 30 women also to tetanus) vaccination. The blood samples were collected before and 4 weeks after the antigens exposure. Immunomodulatory factors: participant's age, body fat, and free testosterone level, were controlled. RESULTS: Apart from the weak positive association between CD3 or CD19 and cFA in men, we found no association between the level of body symmetry and the rest of the analyzed immune parameters for both sexes. DISCUSSION: Our results are the opposite of the good genes hypothesis prediction and suggest that in western, healthy populations, human mate preferences for more symmetric bodies are not related to immune competence.

Background: Identification of new predictive markers in melanoma is of great clinical importance. This study was aimed to analyze association between selected common variants in the cancer susceptibility genes and melanoma progression at the time of diagnosis. Material and Method: The study included 243 consecutive patients with melanoma. Genotyping was performed using real-time PCR. Results: Our data revealed modest association between xeroderma pigmentosum complementation group D (XPD) codon 312 polymorphism and tumor thickness (as defined by Breslow score; XPD D312N CC: 3.00 ± 3.78mm, CT: 1.71 ± 2.48mm, TT: 2,53 ± 3,24mm, P=0.023). The CT genotype in XPD D312N polymorphism was more frequently represented in non-invasive melanomas compared to deeply penetrating tumors. None of the common SNPs in cyclin dependent kinase inhibitor 2A (CDKN2A), vitamin D receptor (VDR), melanocortin 1 receptor (MC1R) were associated with Breslow depth. Conclusion: These findings suggest that genetic alteration in XPD contributes to melanoma progression and may be a potential diagnostic and molecular prognostic marker.

The observations that testosterone might be immunosuppressive, form the basis for the immunocompetence handicap hypothesis (ICHH). According to ICHH only high-quality individuals can maintain high levels of testosterone and afford the physiological cost of hormone-derived immunosuppression. The animal and human studies that attempted to support the ICHH by precisely defined impairment of immunity associated with high testosterone levels are inconclusive. Furthermore, human studies have used only selected immune functions and varying testosterone fractions. This is the first study examining the relationship between multiple innate and adaptive immunity and serum levels of free testosterone, total testosterone, DHT and DHEA in ninety-seven healthy men. Free testosterone and marginally DHT levels were positively correlated with the strength of the influenza post-vaccination response. Total testosterone and DHEA showed no immunomodulatory properties. Our findings did not support ICHH assumptions about immunosuppressive function of androgens. In the affluent society studied here, men with higher levels of free testosterone could afford to invest more in adaptive immunity. Since the hormone-immune relationship is complex and may depend on multiple factors, including access to food resources, androgens should be treated as immunomodulators rather than implicit immunosuppressants.

According to the good genes hypothesis and energy allocation theory, human adult body height may reflect biological quality. An important aspect of this quality is immune system functioning (ISF). The aim of this study was to evaluate the relationship between ISF and body height in healthy people. The ISF was determined by several important innate (total complement and lysozyme activity, neutrophil function) and adaptive immune parameters (lymphocytes, IgA and IgG, and response to the flu vaccine). Overall, 96 males and 97 females were subjected to flu vaccination, and of these, 35 males and 34 females were subjected to tetanus. Blood samples were collected before and four weeks after vaccination. Immunomodulatory factors, participant's age, body fat, and free testosterone levels, were controlled. There was no association between body height and all analysed immune parameters for both sexes. That might suggest that in Western society, a women's preference for taller men is not related to 'good genes for immune competence'. We propose the novel Immunity Priority Hypothesis that explains the lack of relationship between adult body stature and ISF. This hypothesis, however, does not contradict the signalling role of a man's body height as a morphological marker of biological quality.

BACKGROUND: Male gender and adiposity are considered to be risk factors for Staphylococcus aureus carriage. We tested whether colonization is related to free testosterone (fT) level and adiposity, measured with body mass index (BMI) and body fat percentage (BFP), in healthy adults. METHODS: Blood sample and throat swabs were taken twice (at 4-week intervals) from healthy men and women aged 18-36 years. fT level, height, weight, and BFP were measured. Participants were classified as persistent carriers, intermittent carriers (excluded from the analyses), and noncarriers. The final sample was 152 participants: 85 men and 67 women. RESULTS: BFP, but not BMI, correlated positively with S aureus colonization (P = .02) in men. BMI became a significant predictor of carriage only when comparing groups within and above norms (P = .04). There was no relationship for BMI nor BFP in women. Higher fT level was related to persistent carriage (P = .02) in women, there was no relationship for fT level in men. CONCLUSION: Risk factors for S aureus carriage are sex dependent. Within-sex variation in colonization is related to fT level in women, whereas in men it is related to the amount of body fat.

According to the compensatory prophylaxis hypothesis, women in the luteal phase, characterized by a high progesterone level, which suppresses various mechanisms of immune response, should exhibit higher disgust sensitivity, compared to the follicular phase. In this study we test the hypothesis on the compensatory role of disgust sensitivity at the luteal phase of the menstrual cycle, when immune functions are expected to change due to a rise in progesterone level. Disgust sensitivity, progesterone level (P) and white blood cell count (WBC), a general marker of immunocompetence, were measured in 30 healthy women of reproductive age. Disgust sensitivity was evaluated with: 1) Disgust Scale Revised (DS-R) containing three subscales: Core Disgust, Animal Reminder and Contamination Disgust, 2) Pathogen Disgust and Moral Disgust domains of the Three-Domain Disgust Scale. Measurements were conducted twice - in menstruation (the lowest P) and in the mid-luteal phase (the highest P). The results were analyzed longitudinally and using cross-sectional comparisons. Progesterone level, WBC count, and the level of disgust sensitivity in Animal Domain were higher in the mid-luteal phase comparing to menstruation. The level of disgust sensitivity (DS-R score, Animal, Contamination, Pathogen Disgust) correlated only with P (not WBC) and only in the mid-luteal phase (not in menstruation) in between-subjects comparisons. On the base of these results, we hypothesize that the level of disgust sensitivity in the whole menstrual cycle of a woman is "adjusted" to the luteal phase with the highest P level i.e. when immunosuppression is the greatest.

OBJECTIVES: Total leukocyte count (white blood cells-WBC) and the count of each subpopulation vary across the menstrual cycle, but results of studies examining the time and direction of these changes are inconsistent and methodologically flawed. Besides, no previous study focused on leukocyte count on the day of ovulation. METHODS: Blood samples were obtained from 37 healthy and regularly cycling women aged 19.8-36.1 years. Samples were taken three times: during menstruation (M), ovulation (O), and in the mid-luteal phase (ML). WBC, neutrophils, lymphocytes, mixed cells, progesterone (P,) and estradiol (E) were measured in each of the three target phases of the cycle. RESULTS: Compared to menstruation, WBC (P = 0.002) and neutrophils (P < 0.001) increased around ovulation and remained stable in the mid-luteal phase, whereas lymphocyte and mixed cell counts did not change throughout the menstrual cycle. There were some correlations of sex hormone variation with leukocyte changes between M and O (positive for E and WBC, negative for P and WBC and for P and neutrophil count; P < 0.05), but not between O and ML. CONCLUSIONS: Peripheral leukocyte changes taking place in the second half of the cycle are already observable on the day of ovulation and they are associated with sex hormone variation. We speculate that these changes may lead to increased immune protection against pathogens at a time when fertilization and implantation typically occur. Am. J. Hum. BACKGROUND AND AIMS: Epigenetic regulation plays an important role in the management of plant growth, development and response to stress factors, and several reports have indicated that DNA methylation plays a critical role in seed development and viability. This study examines changes in 5-methylcytosine (m(5)C) levels in the DNA of seeds during ageing, a process that has important implications for plant conservation and agriculture. METHODS: Changes in the global level of m(5)C were measured in mature seeds of oak, Quercus robur. The extent of DNA methylation was measured using a protocol based on two-dimensional thin-layer chromatography. Viability of seeds was determined by germination and seedling emergence tests. KEY RESULTS: An ageing-related decrease in total m(5)C during storage of recalcitrant seeds was highly and significantly correlated with a decrease in seed viability, as reflected by a reduction in germination (r = 0·8880) and seedling emergence (r = 0·8269). CONCLUSIONS: The decrease in viability during ageing of Q. robur seeds is highly correlated with a global decline in the amount of m(5)C in genomic DNA, and it is possible that this may represent a typical response to ageing and senescence in recalcitrant seeds. Potential mechanisms that drive changes in genomic DNA methylation during ageing are discussed, together with their implications for seed viability.

5-methylcytosine, an abundant epigenetic mark, plays an important role in the regulation of plant growth and development, but there is little information about stress-induced changes in DNA methylation in seeds. In the present study, changes in a global level of m5C were measured in orthodox seeds of Acer platanoides L. during seed desiccation from a WC of 1.04 to 0.05-0.06 g H2O g g(-1) dry mass (g g(-1)). Changes in the level of DNA methylation were measured using 2D TLC e based method. Quality of desiccated seeds was examined by germination and seedling emergence tests. Global m5C content (R2)increase was observed in embryonic axes isolated from seeds collected at a high WC of 1.04 g g(-1) after their desiccation to significantly lower WC of 0.17 and 0.19 g g(-1). Further desiccation of these seeds to a WC of 0.06 g g(-1), however, resulted in a significant DNA demethylation to R2 » 11.52-12.22%. Similar m5C decrease was observed in seeds which undergo maturation drying on the tree and had four times lower initial WC of 0.27 g g(-1) at the time of harvest, as they were dried to a WC of 0.05 g g(-1). These data confirm that desiccation induces changes in seed m5C levels. Results were validated by seed lots derived from tree different A. platanoides provenances. It is plausible that sine wave-like alterations in m5C amount may represent a specific response of orthodox seeds to drying and play a relevant role in desiccation tolerance in seeds.

Zahavi's handicap principle suggests that only organisms with good genetic quality can afford to engage in costly behaviors. Recreational drug use can be harmful to one's health and therefore might be viewed as a costly signal of one's genetic quality. One of the measurements of genetic quality is bodily symmetry assessed by fluctuating asymmetry. If unhealthy drug use is a behavioral example of Zahavi's handicap principle, then men who use different stimulants or recreational drugs should be more symmetrical than men who do not use them at all or use them only in low quantity. The aim of this study was to examine the relationships between drug use and fluctuating asymmetry. The subjects were 190 young women and 202 young men. Six bilaterally symmetrical traits were measured: length of II-V digits, wrist breadth, and ear height. Questionnaires included questions about smoking, alcohol drinking, drug use, and designer drug use. There was no relationship between bodily symmetry and smoking frequency, alcohol drinking frequency, drug or designer drug use, total substance use, age of smoking initiation, or reason of this initiation. The results indicate that drug use does not reflect genetic quality and does not necessarily relate to the handicap hypothesis.

OBJECTIVE: Body height, body mass index (BMI) and waist-to-hip ratio (WHR) are the main traits characterizing human body morphology. Studies show that these traits are related to attractiveness and, therefore, according to an evolutionary point of view, are supposed to be honest signals of biological quality. If the immunocompetence handicap principal (IHP) is true, people with more attractive values of these traits should be more immunologically competent. To test this, we analyzed whether nasal and throat colonization with potentially pathogenic bacteria is related to body height and BMI in both sexes and to WHR in females. METHODS: 103 healthy females and 90 healthy males (with the mean age of 21.4 and 22.8, respectively) participated in the study. The heights and weights were self-reported and waist and hip circumferences measured. Six potentially pathogenic species (with the most common Staphylococcus aureus) isolated from nasal and throat swabs were identified by colony morphology, standard biochemical assays, and latex tests. To compare carrier and noncarrier individuals, Kruskal-Wallis test was used. RESULTS: Colonized males had higher BMI than non-colonized males (no difference for females) and colonized females had lower WHR's. Body height was not related to colonization in either sex. CONCLUSIONS: We confirmed our hypothesis only for BMI in males. This result and a higher WHR in non-colonized females indicate higher immunocompetence of those who bear the costs of higher levels of testosterone, which according to previous studies is correlated negatively to BMI in males and positively to WHR in females.
